A. Skin Puncture for Adults
1. Disinfect the area with 70% isopropyl alcohol.
2. Allow the site to air-dry.
3. Puncture the preferred site with a sterile disposable lancet. The
puncture is ideally 2 – 3 mm in depth.
4. Wipe off the first drop of blood and collect the subsequent
drops. Avoid squeezing the puncture site, as this will alter the
composition of the blood.
5. Place a cotton ball or gauze on the collection site and instruct
the patient to apply pressure.
6. Discard all used materials properly and wash hands.

B. Syringe Method
1. Apply the tourniquet 3 - 4 inches above the puncture site in a
half-knot manner to congest the vein.
2. Cleanse the puncture site with 70% isopropyl alcohol and allow
it to dry.
3. Puncture the vein with a sterile 20 – 21 - gauge needle attached
to a syringe at a 15°angle and a depth of about 0.5 - 1 cm.
4. Loosen the tourniquet and gently aspirate the blood.
5. After obtaining the required quantity of blood, place sterile
gauze or cotton over the punctured site then withdraw the
needle.
6. Apply pressure to stop bleeding. Never flex the arm.
NOTE: If bleeding is not arrested, elevate the arm and apply more
pressure.
7. Detach the needle from the syringe and transfer the blood
along the side of an appropriate test tube. If the tube is
anticoagulated, mix the tube with blood by gentle inversion
and label the tubes appropriately.
8. Discard all used materials properly and wash hands.

Procedure: HEMATOCRIT DETERMINATION
A. Adam’s Microhematocrit Method
1. Let the capillary or well-anticoagulated blood (from Blood
Collection Procedures) to enter the capillary tube until they are
three-fourths full. Prepare 2 samples. Avoid air bubbles.
2. Seal one end of microhematocrit tubes with clay. Make sure
that the blood is not forced out of the tube during sealing.
3. Place the microhematocrit tubes in the radial grooves of the
centrifuge head with the sealed end away from the center.
4. Centrifuge for 5 minutes at 10,000 G – 15,000 G.
5. Remove the microhematocrit tubes and read on the
microhematocrit tube reader.
6. Results of the two tubes should agree within 2% of the
hematocrit result.
7. Discard used materials properly and wash hands.
