EXPERIMENT : 8

DNA EXTRACTION.

AIM: To extract DNA from the given plant material.

MATERIALS REQUIRED :

Fruits (Banana , Kiwi) , water,,alcohol able salt, zip seal plastic bag., strainer, tooth pick, dish soap
etc.

PROCEDURE :

I. Place the strawberry in a zip seal plastic bag. Press the air out and seal it.
II. Gently mash the bagged strawberry with your fingers for 5 minutes the bag

Once it is turned into a paste, set the bag aside.

III. .Fill a clear glass with half a cup of water slowly add 2teaspoon of dish soap and half table
spoon of table salt. Gently mix this solution without forming bubbles until the salt dissolves.

Add this solution to the plastic bag containing the squashed fruit and mix it carefully

IV. .Flatten out the bag to remove most of the air and then seal it up.
V. Let this mixture remain for 10-20 minutes to allow the soap to release a lot of DNA.
VI. .Place strainer on top of beaker and pour fruit mixture into it. This solution contains
DNA.

VII. .Since DNA is insoluble in water we can use alcohol to precipitate DNA from solution.
VIII. Take some solution in a test tube and slowly drizzle ice-cold ethanol along the side of the test
tube until DNA begins to appear. It will look somewhat like cotton candy fibers.

The DNA will come out of the solution at the boundary layer between alcohol and water which
appear as white shiney strings

viii.We can collect DNA using tooth pick.

OBSERVATION:

DNA appear at the junction of alcohol and water as white shiney strings.

PRECAUTION:

 Mix carefully, the fewer bubbles created the better your result
 Tilt the test tube and very slowly pour the alcohol down its side
 Do not let the banana liquid and alcohol mix.
