SIMMONS CITRATE AGAR RESULTS

Test tubes with 4ml each citrate Test tubes with citrate after seeding of bacteria and
(without inoculating bacteria). incubation at 37°C (24-72 hours)

Positive: Bacterial growth with an intense blue Negative: The culture medium remains green, only a
color is observed in the flute beak slight gas production is distinguished.
INTERPRETATION:

Once the incubation period of 24-72 hours is completed, taking into account that there are
some bacteria that need up to 7 days of incubation.

It is possible to observe in the test tubes the development of colonies on the surface of the
agar and the change in pH, which is distinguished by the change from green to a strong
blue. In this case two results were observed, in that the positive result is the
microorganisms that can use the citrate as a carbon source, presenting a development, it is
this medium that will change to blue. On the other hand, in the negative result no changes
in pH will be distinguished, however, minimal gas production is observed.

A very important detail is that the evaluation of the results would be acceptable as long as
the time and temperature conditions are appropriate. Therefore, prolonged incubation
periods or higher temperatures could alter culture results.

In this case, the possible microorganisms that
could be in this culture medium are Pseudomonas aeruginosa and Escherichia coli due to
the characteristics of the bacteria itself, but at the same time taking into account the soil
from which they were isolated.

DISCUSSION OF RESULT

bacteria from a contaminated soil, just like our research One of those tests was Simmons citrate to
determine gram-negative bacteria and this tested positive for the bacteria Pseudomonas aeruginosa,
but the difference is that it confirmed through a molecular identification test and thus the colonies
were able to be distinguished. And this was be an excellent degrader of petroleum hydrocarbons, so
more specialized studies should be carried out because it is an important environmental problem.
