SECTION 11: Toxicological Information - For ZDEC Powder (14324-55-1)
Harmful if swallowed.
Oral LD50 > 1960mg/kg (rat)
ZDEC can however cause irritation and soreness to skin and eyes with long term exposure.
Toxicokinetics, metabolism and distribution Toxicokinetic data are available on the structural analogue of ZDEC, ZDMC. Based
on the higher molecular weight of ZDEC, it is expected that its absorption from the gut shall not be faster than that of ZDMC. For
ZDMC, at least 60% oral absorption was determined based on the available toxicokinetic study. The distribution of ZDEC,
similarly to ZDMC, is expected to occur primarily to organs of metabolism and excretion (liver, lung, kidney) and vascularised
tissues (spleen, thyroid, adrenals), as well as body fat. The metabolism is expected to occur via a hydrolysis of a parent compound
into the respective acid, which either undergoes a transformation to CS2, further oxidized into CO2, or a conjugation with
glucuronic acid or GSH. For the dermal absorption, a value of 0.1% as observed in the study with ZDMC is considered to be a
worst-case approach (due to the very low solubility of ZDEC and the possible amount of water that can penetrate the skin,
0.00012% would be observed maximally).
CMR effects (carcinogenity, mutagenicity and toxicity for reproduction)
- Genetic toxicity:
in vitro (mamalian cell gene mutation assay, mouse lymphoma cells) - negative
in vitro (chromosome abberation, human lymphocytes) - positive
in vivo (rats, unscheduled DNA synthesis) - ambiguous results
in vivo (mouse, chromosome abberation) - negative
- Carcinogenicity : NOAEL: 717 mg/kg bw/day
- Reproductive toxicity: effects on fertility : NOAEL (maternal): 10 mg/kg bw/day NOAEL (neonatal): 10 mg/kg bw/ day (Read
Across from ZDMC (CAS 137-30-4)
toxicity on development : NOAEL (maternal): 125 mg/kg bw/day 250 mg/kg bw/day
May cause sensitisation by skin contact
