J. van Oeveren et al. (2011). Sequence-based physical mapping of complex genomes by Whole Genome Pro ling. Genome Research 21: 618-625 www.wholegenomepro ling.com
Examples of genomes with WGP aided assembly - Melon (0.4 Gb) - Lettuce (2.6 Gb) highly repetitive - Sun ower (3.6 Gb) large & repetitive - Tobacco (4.6 Gb) tetraploid - Wheat Chromosome by chromosome Now the WGP technology is more affordable than ever by making the technology compatible with the Illumina Hi-Seq 2000 platform, unprecedented data production at signi cantly reduced cost became possible. WGP is offered at three different application levels: - WGP-gold based on 12x BAC library coverage - WGP-standard based on 10x BAC library coverage - WGP-annotated based on 2-6x BAC library coverage
Prof. Rod Wing Arizona Genome Institute I am a strong proponent of using physical maps as backbones for the generation of new reference genomes. Several genomes published without the use of physical maps have more than half the genome missing. Callum Bell National Center for Genome Resources KeyGenes method offers a physical map in a shorter amount of time and at a lower cost compared to other methods. Physical maps go a long way in building bigger scaffolds.
Michiel van Eijk KeyGene To date, we have completed high-quality WGP maps for over 20 crops ranging in genome size between 130 Mbp and 5 Gbp, including highly repetitive diploids such as lettuce and pepper and polyploids such as canola and tobacco. Our results demonstrate unequivocally the versatility of WGP to deliver superior sequence-based physical maps irrespective of genome size and/or repeat content.
