Paper 2: The Influence of pH on Enzyme Activity in Catalase

Abstract
This study examines how pH affects the activity of the enzyme catalase. By testing catalase
activity at different pH levels, we determined that catalase is most active at a pH of 7, with
activity decreasing at higher and lower pH levels.

Introduction
Enzymes are proteins that catalyze biochemical reactions. Their activity is highly dependent on
environmental conditions, including pH. Catalase, an enzyme found in many living organisms,
decomposes hydrogen peroxide into water and oxygen. This experiment investigates the
optimal pH for catalase activity.

Methodology

● Materials: Catalase solution, hydrogen peroxide, pH buffers, test tubes, stopwatch.


● Procedure: Measure the rate of oxygen production by catalase at pH levels of 4, 5, 6, 7,
8, and 9. Use pH buffers to maintain the desired pH.
● Data Collection: Record the volume of oxygen produced in a fixed time period.

Results

● pH 4: Low activity
● pH 5: Moderate activity
● pH 6: High activity
● pH 7: Highest activity
● pH 8: Moderate activity
● pH 9: Low activity

Discussion
Catalase shows highest activity at pH 7, suggesting it operates best under neutral conditions.
Extreme pH levels likely denature the enzyme, reducing its activity.

Conclusion
The optimal pH for catalase activity is around 7. Deviation from this pH results in decreased
enzymatic activity, highlighting the importance of maintaining proper pH levels for enzyme
function.
