blindness. When handling chemicals, always check the safety data sheet
and wear protectors, such as safety goggles and safety gloves.

WARNING Handling of septum, washing

solution, and waste solution
Change the septum for the measuring vials and 28 mL vial with a new
one every measurement day. Replace the washing solution with a new
one every measurement day.
Dispose of waste solution collected in the 28 mL vial every measurement
day. If waste solution is over vial capacity, it will overflow on the unit and
a fire may be caused.

POINT Liquid volume in the vial

If the liquid volume in the measuring vials is small, the needle tip of the
gastight microsyringe may not reach and not be able to aspirate the
liquid. In addition, the volume may decrease during repeated
measurement and the liquid may not be aspirated properly.

POINT Liquid contamination

If the measuring vial or septum to use is contaminated, the liquid to be
measured may be contaminated. If the liquid to be measured is low-
concentration, rinse the vial two to three times with the septum and cap
on. To avoid the contact between the liquid and septum, the liquid volume
should be up to 80% of the measuring vial.

ASC-550L (NSX-5000)
Measurement 5-3

(1) Pour a calibration curve solution or sample in the measuring vials up to 80% of the
vial volume. Set them in the vial position for measurement from 1 to 50 or 1 to 105.
For the calibration curve solution, refer to each measurement item of the NSX
series calibration curve stock solution. Prepare the stock solution for the
calibration curve and dilute it with an optimum solvent.

(2) Pour a washing solution into a 28 mL vial and set it to A, vial position for washing
solution. For the washing solution, add an optimum organic solvent for
nonaqueous sample measurement, and pure water for aqueous sample
measurement.

(3) Prepare a 28 mL vial that match the functions set for Btl. B. Set it to B, vial
position for multipurpose. For the setting details of Btl. B, refer to “5-3.
Parameters”.

(4) Set an empty 28 mL vial to W, vial position for waste solution.

unit. For the setting of vial tray, refer to “3-6. Setting of Vial Tray”.

5-2. Operation after Measurement

5-2-1. Washing of Gastight Microsyringe

After measurement, wash a gastight microsyringe and store it as follows. When it is left
unwashed, the plunger adheres to it and load is applied on the ASC-550L unit at
plunger operation. It may cause a malfunction.

WARNING For protection

When handling chemicals, always wear protectors.
Failure to do so may lead to physical damage, such as chemical injury or
blindness. When handling chemicals, always check the safety data sheet
and wear protectors, such as safety goggles and safety gloves.

(1) Remove the gastight microsyringe from the syringe holder.

(2) Aspirate and inject a solvent that dissolves the measured sample well to wash the
inside of the gastight microsyringe. If the tip of the plunger is contaminated,
remove the plunger and wipe it off with a waste cloth.

(3) Aspirate and inject alcohol. Then, aspirtate and inject air to dry the inside.

(4) To protect the needle, store the gastight microsyringe in a case.

Details of ASC Program

(1) Sampling
Perform setting according to the liquid properties to measure. Especially when the
viscosity is high, slow down the aspiration rate or increase the waiting time after
aspiration. For the order of sampling to the gastight microsyringe, refer to “Fig. 5-2.
Sampling Order”.

Post Air

Fig. 5-2. Sampling Order

(2) Injection
When oil is selected as the measurement mode in the method file or schedule file
settings, the setting range will be low speed. When aqueous sample is selected, it
will be high speed. If the injection rate is high, incomplete combustion may occur.
Basically, do not set the rate faster than the default value.

(3) Washing
Set the washing rate, volume and number of times. The washing solution volume
is set as a percentage (%) of the gastight microsyringe volume.

(4) Rinse
Set the rinse rate, volume and number of times. The rinse volume is set as a
percentage (%) of the gastight microsyringe volume. A liquid used for rinse
depends on the Bottle B setting. When Bottle B is set to Solvent Flush, rinse with
the liquid in Bottle B. For other settings, rinse with a liquid to measure.

5-4. Automatic Syringe Dilution

With ASC-550L, by sampling a solvent put in any measuring vials together with a
sample liquid, they can be automatically diluted and measured in the gastight
microsyringe. As the solvent to be diluted, select a solvent that can sufficiently dissolve
the sample liquid. For details on the setting method of automatic syringe dilution, refer
to the operation manual.
