STROMAL BESTOWER

Stem cell, in living organisms, an undifferentiated cell that can produce other cells that
eventually make up specialized tissues and organs. There are two major types of stem
cells, embryonic and adult. Embryonic stem cells are located in the inner mass of a
blastocyst (an embryo at a very early stage of development), and they eventually give
rise to every cell type of the adult organism. In cancer treatment, blood-forming adult
stem cells are routinely harvested from bone marrow, stored, and then reinfused into
patients to replace blood cells destroyed by chemotherapy or radiation therapy. This
potential for replacing damaged tissues has aroused great interest in using embryonic
stem cells to treat a number of other conditions, such as Parkinson disease, severe burns,
and damage to the spinal cord. However, there is a demand in donating the stemcell and
finding it for a proper individual. If there would be place like a stem cell institution,
there were of money minded in such case this fails here. So, in that casethis project will
provide a solution on finding the perfect donor by the seeker itself through a website
mode by which both the donor and the seeker encounter with each self-alongside,
furthermore their intensions will become fruitful.

1. PROJECT REJUVENATION AND REFINEMENT PLANNING

1.1 Introduction:
Stem cells are the body's raw materials cells from which all other cells with
specialized functions are generated. Under the right conditions in the body or a
laboratory, stem cells divide to form more cells called daughter cells. These daughter
cells become either new stem cells or specialized cells (differentiation) with a more
specific function, such as blood cells, brain cells, heart muscle cells or bone cells.
No other cell in the body has the natural ability to generate new cell types. In cancer
treatment, blood-forming adult stem cells are routinely harvested from bone marrow,
stored, and then reinfused into patients to replace blood cells destroyed by
chemotherapy or radiation therapy. This potential for replacing damaged tissues has
aroused great interest in using embryonic stem cells to treat a number of other
conditions, such as Parkinson disease, severe burns, and damage to the spinal cord.
Though it has it own advantages, there is a great demand on find the proper donor
for the patient. And hence this project will find a direct solution on finding its donor
by the patient itself. This convey that it can only aid and enhance our lives if we get
on board with technological advances, and also will pay a way at the forefront.

creating a website where their will be donation option and their will be a portal for
the seeker to find their correct match by oneself. Usability requirements are the
things your website users need to complete their goals a leave satisfied. Basic
usability requirements include clarity, consistency, and accessibility. It is all met
here in this project through which the patient itself can find their appropriate donor.
1.2 Objective:

Stem cells are at the forefront of one of the most fascinating and revolutionary areas
of medicine today. Doctors recognise that stem cells have the potential to help treat
numerous diseases by generating healthy new cells and tissue. There are a wide range
of diseases that are treatable with stem cells derived from cord blood and other
sources of similar type of stem cells (Hematopoietic Stem Cell), like bone marrow
and peripheral blood, including stem cell disorders, acute and chronic forms of
leukemia, myelo proliferative disorders, and many more. In addition to the host of
conditions that can now be treated, it is the potential of stem cell treatments that
holds the most excitement as research continues to uncover new possibilities. The

● Interface: In this section, the admin can view all the details in brief like the
total stem cells listed, the registered donor list, and the total inquiries
received.
● Database Management: In this section, the admin will manage all the
information that has been collected from the recipient and a donor.
● Donor List: In this section, the admin can view a list of donors and their
respective information.
● Manage to Contact us Query: In this section, the admin can manage a query
that is received by users.
● Managing pages: In this section, the admin can website pages.

● Updating Contact info: In this section, the admin can update the contact
details of the website.
● Request Acceptance: In this section, the admin can view the request for
blood that the donor receives.

● Donor Source: Users can view and contact donors.
● Seek Fundraiser: Users can look for volunteers through the geographical
area and blood type.
● My Account: Profile, Change Password, Request Received, Logout
Moreover, donors can update their passcode, change their identity, and recover
their passcode. By incorporating these features, the portal not only serves as a
matching platform. The stem cell donation portal transforms into a dynamic and
interactive platform that not only facilitates matches but also creates a vibrant and
supportive community dedicated to making a meaningful impact in the realm of
healthcare and patient well-being.

The flowchart for registered users on the stem cell donation portal, as depicted in the
encompasses user-friendly steps such as registration, profile creation, exploration of
the matching algorithm, active searching, secure communication, community
engagement, volunteer participation, educational resource access and recognition.
This user-centric approach ensures that individuals leave the portal satisfied, having
achieved their goals of finding a suitable match or contributing to the cause of stem
cell donation. The database includes user profiles data, matching algorithm data,
educational content data, community forum data, notification data and recognition
data.

2. RESEARCH AND LITERATURE REVIEW

2.1 Depth of Research:

There are three types of cells are collected for stem cell transplant

❖ PERIPHERAL BLOOD STEM CELL TRANSPLANT

❖ BONE MARROW TRANSPLANT

❖ CORD BLOOD TRANSPLANT

-> And hence cord blood type of stem cell transplant is used for the process.

2.2 Relevance:

The papers mostly belong to concepts dealing with right to donate in umbilical blood
banking for stem cell, donor selection and management, unrelated adult stem cell
donor medical suitability. The papers were written by renounced scientists and
researchers. Some papers also deal with medical researches and how to overcome it.
2.3 Identification of Challenges:

The main challenge here would be preclinical studies safety assessments and
interaction between regulatory agencies are essential in this field. Stem cell
treatments are very expensive and it is difficult for developing countries to keep up
with technological advances. Promoting private sector funding, international
collaboration, and exchange research should help resolve most of the challenges in
stem cell therapy research and implementation.

2.4 Patents:

1. The right to donate in umbilical cord blood banking for stem cell therapies.
Abstract
To explore the views of lay and professional stakeholders about the donation of
cord blood. Participants claimed pregnant women had a right to know of the value
of cord blood. This highlighted the flaws of the existing donation infrastructure,
which was portrayed as playing a significant role in determining public health.
Participants called for a right to donate cord blood to readdress the inequity in
healthcare services for pregnant women and transplant recipients. Umbilical cord
blood, rich in hematopoietic stem cells, has emerged as a valuable resource for
treating a variety of diseases, including blood disorders, immune deficiencies, and
certain cancers. Donors maintained a sense of right over their donation when they
discussed cord blood donation as potentially benefiting their family as well as
society. In order to keep receiving donated body parts, tissue and blood, there is a
need to take into account the way in which donation operates within a prevalent
'rights' discourse.
2. Unrelated adult stem cell donor medical suitability
Abstract
The World Marrow Donor Association (WMDA) fosters collaboration between
international registries to facilitate the exchange of hematopoietic stem cell
products for unrelated stem cell donor transplantation. As indications for
hematopoietic SCT grow, the movement of products across the world will
increase. There is a need to protect the best interests of donors and recipients by
identifying universal donor medical suitability criteria. Within this report the
WMDA provides a background to unrelated adult donor and recipient safety,
recommends a common framework for assessing the health of unrelated adult
donors at each stage of the donation pathway and presents a novel mechanism for
sharing international consensus criteria for individual medical and lifestyle
conditions.

3. Donor selection and management


Abstract
Impact of HLA diversity on donor selection in organ and stem cell transplantation.
The human major histocompatibility complex is a multigene system encoding
polymorphic human leucocyte antigens (HLA) that present peptides derived from
pathogens to the immune system. The high diversity of HLA alleles and
haplotypes in the worldwide populations represents a major barrier to organ and
allogeneic hematopoietic stem cell transplantation, because HLA
incompatibilities are efficiently recognized by T and B lymphocytes. In organ
transplantation, pre-transplant anti-HLA antibodies need to be taken into account
for organ allocation.
4. HLA Match Likelihoods for Hematopoietic Stem-Cell Grafts
Abstract
Hematopoietic stem-cell transplantation (HSCT) is a potentially lifesaving
therapy for several blood cancers and other diseases. For patients without a
suitable related HLA-matched donor, unrelated-donor registries of adult
volunteers and banked umbilical cord–blood units, such as the Be the Match
Registry operated by the National Marrow Donor Program (NMDP), provide
potential sources of donors. Racial and ethnic groups to predict the likelihood of
identifying a suitable donor (either an adult donor or a cord-blood unit) for patients
in each group. The models incorporated the degree of HLA matching, adult-donor
availability (i.e., ability to donate), and cord-blood-unit cell dose.
5. Allogeneic hematopoietic stem cell transplantation to cure sickle cell disease.
Abstract:
Sickle cell disease (SCD) had first been mentioned in the literature a century ago.
Advancement in the molecular basis of the pathophysiology of the disease opens
the door for various therapeutic options. Though life-extending treatments are
available for treating patients with SCD, allogeneic hematopoietic stem cell
transplantation (HSCT) is the only option as of yet. A major obstacle before HSCT
to cure patients with SCD is the availability of donors. Matched sibling donors are
available only for a small percentage of patients. To expand the donor pool,
different contrasting approaches of allogeneic HSCT like T-cell replete and
deplete have been tested. None of those tested approaches have been without the
risk of GvHD and graft rejection. The center of interest is using mega dose T-cell-
depleted bone marrow in conjugation with donor-derived CD8 veto T cells to
achieve engraftment and tolerance across MHC barriers, under reduced intensity
conditioning (RIC). This approach is in phase I/II clinical trial at the MD
Anderson Cancer Centre and is open to patients with hemoglobinopathies.

i. Stem Cell Identification and Classification:

Technologies related to the identification and classification of different types of stem


cells, including mature and adolescent stem cells. This may involve advanced
techniques for studying cell biology and differentiation. Stem cells are unique cells
with the remarkable ability to develop into various cell types within the body.
Identifying and classifying stem cells is crucial for understanding their potential
applications in regenerative medicine, research, and therapeutic interventions. Stem
cells are undifferentiated cells with the remarkable ability to self-renew and
differentiate into various specialized cell types, making them invaluable for
therapeutic applications and understanding developmental processes. Stem Cells are
identified based on the Morphological Characteristics such as Embryonic Stem Cells
(ESCs), Adult Stem Cells (ASCs).
ii. Cryopreservation Techniques:

Cryopreservation is a vital technique in stem cell research and regenerative


medicine, allowing cells to be stored at extremely low temperatures to maintain their
viability and functionality for future use. Technologies related to the preservation of
blood-forming adult stem cells, which are frequently removed from the bone marrow
during cancer treatment. Cryopreservation involves freezing and storing cells for
later use. Cryoprotectants are substances added to cells before freezing to protect
them from ice formation and damage. Common cryoprotectants include dimethyl
sulfoxide (DMSO) and glycerol. Stem cells are stored at ultra-low temperatures,
typically in liquid nitrogen at around -196°C (-321°F). These extreme temperatures
halt biological activity and preserve cell integrity. As technology evolves, ongoing
improvements in cryopreservation methods will continue to play a pivotal role in
advancing stem cell therapies and regenerative medicine.

iii. Online Portal and Website Development:

Technologies associated with the development of a user-friendly website and online


portal. This includes web development tools, database management, and user
interface design to create a platform where donors and seekers can connect. Employ
web development technologies to create a user-friendly website and online portal.
This includes frontend and backend development, database management, and secure
data handling.The frontend is the user interface that donors and seekers interact with.
It includes the design, layout, and functionality visible to users. The technologies
used in frontend development are CSS, HTML, JAVASCRIPT. The backend is the
server-side of the application, managing data, user authentication, and business
logic. The technologies used in frontend development are APIs (Application
Programming Interfaces), Python.

iv. Matching Algorithm:

Proprietary algorithms or methodologies used to determine compatibility between


stem cell donors and seekers. This technology is crucial for efficiently matching
donors with seekers based on criteria such as tissue compatibility, blood type, and
other relevant medical factors.The algorithm employs sophisticated techniques to
assess the compatibility of potential donors and seekers at the tissue level. This
involves analyzing genetic markers and other molecular characteristics to identify
optimal matches. Special emphasis is placed on evaluating HLA (human leukocyte
antigen) compatibility, a key determinant for successful stem cell
transplantation.The matching algorithm begins by collecting detailed information
from both donors and seekers, including their medical histories, genetic profiles, and
preferences for donation. Donor profiles may include data such as age, gender, HLA
typing, and tissue compatibility, while seeker profiles may include information about
their medical condition, treatment requirements, and urgency of need.

v. Secure and Private Messaging System:

Technologies related to the implementation of a secure and private messaging


system within the website. This involves encryption and secure communication
protocols to protect the confidentiality of the interaction9 between donors and
