ÖSSZEFOGLALÁS
ABSTRACT
Recently, there has been an increasing interest in the use of fermented feeds, as this type
of feed ingredients can have a beneficial effect on the welfare of poultry. Interleukins are
a group of signaling proteins (cytokines) that play a critical role in the immune system.
Interleukins can have both pro-inflammatory and anti-inflammatory effects. In the present
study, IL-6 and IL-10 expression in the small intestine (duodenum, jejunum, ileum) and
caecum was investigated in two groups (control and experimental). Three samples were
taken from the small intestine (duodenum, jejunum, ileum) and caeca of five birds (n =
10) per experimental group. IL-6 expression was significantly (p<0.05) different between

the control and experimental groups in the duodenum, jejunum and caecum. IL-10
showed significant difference at p<0.001 level in the ileum and caeca sections.

Mintavétel
A kontroll és a fermentált takarmányt fogyasztó kísérleti csoport egyedeitől (összesen
10 brojler) szövetmintát gyűjtöttünk a vágás napján (42. nap), a vékonybél egyes
szakaszairól (duodenum, jejunum, ileum, caecum) RNAlater (Thermo Fisher, USA)
oldatban (összesen 40 minta).
RNS izolálás
1,5 ml-es Eppendorf csőbe 1000 µl TRIzolt (Thermo Fisher Scientific, USA)
pipettáztunk, majd 300 mg szövetmintát helyeztünk az oldatba. A mintákat TissueLyser
LT (Qiagen, Németország) segítségével homogenizáltuk 2 mm-es Bashing Beads (Zymo
Research) gyöngyökkel ellátott csövekben. A homogenizált mintát 10 percen keresztül
centrifugáltuk (12000 x g), majd az RNS-t is tartalmazó rózsaszín felülúszót új csőbe
helyeztük. Rövid (5 perc) inkubálást követően 200 µl BCP-t adagoltunk hozzá, majd
Vortex-szel (Velp Scientifica, Olaszország) elegyítettük. Ezt követően 15 percen
keresztül (12000 x g) centrifugáltuk a mintát, majd a színtelen, RNS-t is tartalmazó

felülúszót (kb. 600 µl) új kémcsőbe pipettáztuk. 500 µl izopropanol adagolását követően
óvatosan többször elegyítettük a mintát. A kicsapódás során keletkező RNS-t
centrifugálással pelletáltuk a mintacső aljára. Centrifugálást (12000 x g) követően a
felülúszót eltávolítottuk. 500 µl 70% (V/V) etanolt (DEPC vízben hígítva) adagoltunk a
pelletre. Ismételt centrifugálást követően (10 perc, 12 000 x g) pipettával eltávolítotottuk
a felülúszót. 50 µl nukleázmentes vizet adagoltunk a pelletre.
Az RNS-koncentrációt NanoDrop 2000 spektrofotométerrel értékeltük. A minták
integritását agaróz gélelektroforézissel ellenőriztük. Mintánként 1-1 µg teljes RNS-t
használtunk a DNase kezeléshez (2. táblázat), amelyet a reverz transzkripció követett
RevertAid First Strand cDNA Synthesis Kit (Thermo Fisher Scientific) és oligo dT
primerek felhasználásával (3. táblázat).

Statisztikai értékelés
A normalizált génexpressziós értékeket a kontroll és a kísérleti csoport átlagában vetettük
össze egymással SPSS v.20.0 for Windows statisztikai szoftverben független mintás t-
próba segítségével. A csoportok közötti különbségeket p<0,05 szinten tekintettük
szignifikánsnak.
