Controlled Release Technology and Design of Oral Controlled Released Dosage Forms Design and Scale Up of Drag Granulation Process
Introduction:
An oral controlled release d rug deliver y system is designed to
deliver a drug in a controlled and predictable manner over a period
of time or at a predetermined position in the gastrointestinal tract .
There are several other terms used interchangeably to describe
controlled release dosage forms .
The U.S. Food and Drug Administration denes modied release dosage
forms as those whose drug release characteristics of time course
and/or location are chosen to accomplish therapeutic or
convenience objectives not offered by conventional dosage forms
such as a solution or an immediate release dosage form .
Modied release oral dosage forms include :
1- extended release, that is, dosage forms designed to make the drug
available over an extended period of time after ingestion.
2- Delayed release ,that is ,dosage forms designed to provide a delay
before drug release.
Advantage :
1- Oral dosage forms are preferred because of their convenience and
cost-effectiveness.
2- There are several reasons for pursuing the development of
controlled release dosage forms
3- Controlled release formulations can reduce the dosing frequency
and minimize side effects
4- Orally disintegrating tablets have the advantage that they can be
taken without water This can be very important to pediatric and
geriatric patients, and to patients who have difculty swallowing
tablets or capsules.
