Focusing the Compound Light


Microscope
1. Start with the lowest power objective (10x)
2. Focus using the coarse adjustment (big knob)
3. Switch to the next higher power objective
***When you are on the highest power objective, ONLY
USE THE FINE ADJUSTMENT!!!! Otherwise you could
break the slide!***
As you switch from low to high power, the field of view becomes
darker. To deal with this, the diaphragm needs to be opened to
allow in more light.

4. As you switch from low to high power the field of


view becomes smaller.

Staining
Many parts of the cell are almost
colorless and are hard to distinguish
from other cell parts
We add stain to color cell parts so we
can see them better under the
microscope
Common stains:
Iodine
Methylene blue

Adding Stain (w/o removing


coverslip)

Making a Slide (a wet-mount)

Dissecting Microscope
Allows us to study larger
3D specimens
Ex. Insects
Things that CAN be
seen with the naked eye
2 eyepieces and 2
objective lenses
Helpful in performing
dissections

Electron Microscope
Uses a beam of electrons
instead of light
Focused by magnets
instead of lenses
Disadvantage: can only
use to view dead
specimens
Advantage: very high
magnification allows us to
see internal structure of
cells
