Subjects and Methods

59 healthy young subjects


28 Ala54/Ala54
27 Ala54/Thr54
4 Thr54/Thr54

Completed 3 diets (28 days)


1 high saturated fatty acid (SFA)
2&3 randomized crossover design
Low fat and high carbohydrate diet
High MUFA diet (mediterranean diet)

Blood sampling and biochemical determinations


Venous blood for insulin, glucose, lipid, and lipoprotein collected into EDTA tubes
after 12-h overbight fast at the end of each dietary period
Each analysis was performed 3 times
Total cholesterol and triacylglycerols were assayed by enzymatic procedures
Unesterified free FA enzymatic colorimetric assay
To reduce interassay variation, plasma was stored at 80C and analyze in
duplicate at the end of study

Glucose supression test


Somatostatin was used to inhibit endogenous insulin secretion
SSPI concentration were similar in all subjects, SSPG provided a measure of the
ability of insulin to promote the disposal of infused glucose
Subjects with high SSPG more insulin resistant

Genotyping of FABP2 gene polymorphism


PCR RFLP Assay was developed for genotyping.
The genomic DNA fragment flanking the ALA54/Thr polymorphism was
amplified by using 2 primers flanking exon 2 of the FABP2 gene.

Statistical analysis
Statistical analysis using SPSS version 11.0
The general linear model for repeated procedures was used to test main
effects of genes and diet and gene x diet interactions
When significant effects were observed, Tukeys post hoc test was used to
identify between group differences
Correlation was performed with Pearsons correlation coefficient
