The answers in questions
#3 and #4
Question #3
Most Laboratory media contains a
fermentable carbohydrates and
peptone because the majority of
bacteria requires carbon, nitrogen
and energy source in these forms.
How are these three needs met by
glucose-minimal salt medium?
But…
What is a glucose-minimal
media?
MINIMAL MEDIA
are those that contain the minimum, nutrients
possible for colony growth, generally without the
presence of amino acids , and are often used to grow
wild types of microorganisms.
can be also used to select for or against recombinants
or exconjugants.
usually contains…..
a carbon source for bacterial growth which may be
glucose, or a less energy-rich source such as succinate
various salt which may vary among bacteria species
and growing conditions that generally provide
essential elements such as magnesium, nitrogen,
phosphorus and sulfur to allow bacteria to synthesize
protein and nucleic acid. Ammonium chloride is
commonly used, particularly given that the majority of
the bacterial species cannot fix nitrogen.
and….
Ions or electrolytes- such as
sodium, chloride, potassium,
calcium, phosphorus, and
magnesium are also needed
by the bacteria.
Question #4
Flask A contains yeast cells in glucose-minimal salt
broth incubated at 30 degrees with aeration. Flask B
contains yeast cell in glucose minimal salt broth
incubated at 3o degrees in an anaerobic jar. the yeast
are the facultative anaerobes.
a. Which culture produced more atp?
b. Which culture produced more alcohol?
c. Which culture has a shorter generation time?
d. Which culture has the greater cell mass?
e. Which culture has the higher absorbance?
a. Which culture produced more atp?
flasks A the oxidation of glucose in aerobic respiration
is..
C6H12O6 + 6oxygen + 38 ADP + 38 PHOSPHATE ----
6CO2 +6H20 +38 ATP
Because the facultative anaerobe which uses
fermentation pathway that produced only 2 atp.
formula: glucose + 2ADP + 2 phosphate --- 2 C6H5OH
+ 2 CO2 + 2 ATP.
b. Which culture produced more alcohol?
flask B because
fermentation occur in
the absence of oxygen
produces 2 ethanol
c. Which culture had a shorter generation
time?
bacteria are said to be aerobic f they require O2 and
grown best at higher oxygen tension usually 20 percent
or more, but microaerophilic bacteria needs O2 but
grows best or reduced O2 tension less than 1o percent.
Facultative bacteria that makes ATP by aerobic
respiration if oxygen is present but is also capable of
switching to fermentation, therefore, the culture in
Flask A has a shorter generation time.
d. Which culture has the greater cell mass?

Flask A, because
it has a shorter
generation time.
e. Which culture had the higher
absorbance?
Flask A because it’s
environment is open
where oxygen is
available.
