Microscopic Examination
 The Procedure:
a sample of feces is mixed with saline/ eosin 1% to
form a slurry.
 A drop of this mixture is placed on a glass slide
 Cover with a coverslip
 examined under the microscope

 Observe:
 Meat fiber
 Leucocytes/ erythrocytes
 Bacteria, Cyst, worm egg
Fecal fat examination :
 Procedure:
 Put one drop of acetic acid 36% on objective
glass,
 add a little amount of feces, mix well
 Add with one drop of Sudan III reagent, mix
and boil in open fire, then close with the deck
glass.
 Analyze microscopically, in 40X objective lens
Fecal Occult Blood Test
 Principle:
 Immunoassay for detection of human blood in
feces
 The membrane is precoated with anti-Hb
antibody on the test line
 The specimen migrate upward on the
membrane chromatographically by capillary
action to react with anti-Hb antibody
generated colored line
Procedure
 Unscrew the top of the sample collection device
and use the sample collection stick to collect
stool sample by dipping the stick into 3 different
places of the same stool sample.
 Put back the sample collection stick into same
collection device and screw it tightly. Shake the
collection device vigorously
 Break off the tip of the collection device and
squeeze 2 drops of the extracted sample into the
sample area.
 Read result at 5 to 10 minutes
