DRUG DISTRIBUTION
Distribution is a branch of pharmacokinetics
which describe the reversible transfer of a drug
from one location to another within the body.
Once a drug enters into systemic circulation by
absorption or direct administration ,it must be
distributed into interstitial and intracellular fluids

FACTORS AFFECTING DRUG DISTRIBUTION

Physical volume of an organism:


The differing specificities of different tissue will give rise to different
concentration of the drug within each group.

Removal rate:
A drug removal rate will be determined by the proportion of the drug that is
removed from circulation by each organ.
Plasma protein:
This is important as only drug that are present in the plasma in their
free form can transported to the tissue. Drugs that are bound to plasma
proteins therefore acts as a reservoir of the drug

Physico-chemical properties:
Important physico chemical properties which influenze the drug
distribution is
•Molecular weight
•Degree of ionization
Physiological barriers:
•Simple cell membrane
•Blood brain barrier
•Cerebrospinal spinal fluid barrier

Binding:
The distribution of a drug in the body depends on the extent to which
the drug binds to protein and tissue in the body. Only drugs that are bound to
proteins and other components in the blood are free to diffuse across the cell
membrane into the tissue of the body.
