MUSCLE TRANSDUCES
CHEMICAL ENERGY INTO
MECHANICAL ENERGY
Muscle:
The major biochemical transducer that converts potential energy into
kinetic energy
Largest single tissue in the human body (makes up somewhat less than
25% of body mass at birth, more than 40% in the young adult, and
somewhat less than 30% in the aged adult)
Three types of muscles found in vertebrates: skeletal, cardiac, and
smooth.
Troponin complex
• unique to striated muscle
• accounts for the inhibited state of relaxed striated muscle
• consists of three polypeptides: Troponin T (TpT), Troponin I (TpI),
Troponin C (TpC)
TROPOMYOSIN & THE TROPONIN COMPLEX
PRESENT IN THIN FILAMENTS PERFORM KEY
FUNCTIONS IN STRIATED MUSCLE
1. Troponin T (TpT) - binds to tropomyosin as well as to the other two troponin
components
2. Troponin I (TpI) - inhibits the F-actin-myosin interaction and also binds to the
other components of troponin
Two Isoforms:
• RYR1 - skeletal muscle
• RYR2- heart muscle and brain
Treatment:
Dantrolene - a skeletal muscle relaxant that acts to
inhibit release of Ca2+ from the SR into the
cytosol, thus preventing the increase of cytosolic
Ca2+ found in MH
MUTATIONS IN THE GENE ENCODING THE
CA2+ RELEASE CHANNEL ARE ONE CAUSE
OF HUMAN MALIGNANT HYPERTHERMIA
• CA2+ channels
• Ca2+ in the
myoplasm = Ca2+
release channel
of the SR
• Missense mutations are in the head and headrod regions of the myosin
heavy chain
• Some mutations alter the charge of an amino acid side chain = affects the
conformation of the protein
DILATED
CARDIOMYOPATHY
• Mutations in the genes encoding:
• Dystrophin help organize the contractile
apparatus of cardiac muscle cells
• Muscle LIM protein (Lin-II, Isl-1, and Mec-3)
involved in the regulation of a
• Cyclic response-element binding protein (CREB) number of genes in these cells
• Desmin
• Lamin
When smooth muscle myosin is bound to F-actin in the absence no detectable ATPase
of other muscle proteins such as tropomyosin activity
• The calmodulin-4Ca2+-
activated light chain kinase
phosphorylates the light chains
