Biochemistry Practical 1

(Estimation of Creatinine & Uric acid)

By : Osama Al-Zahrani
Colorimeter
- A colorimeter is a device that measures the absorbance of particular
wavelengths of light by a specific solution.

- This device is commonly used to determine the concentration of a known

solute in a given solution
Uric acid
Estimation
Uric acid
Procedure :
Uric acid
Equation:
Ab.T
Uric acid conc. (mg/dl) = × conc. Standard
Ab.S
Example :
If the Absorption test (Ab.T) = 0.257 ml
If the Absorption standard (Ab.St) = 0.243 ml
If the conc. Standard = 8 mg/dl

0.257
Then : Uric acid conc. = × 8 = 8.46 mg/dl (Hyperuricemia for male & female).
0.243

Normal value of Uric acid in human plasma :

Male : 3.4 – 7 mg/dl
Female : 2.4 – 6 mg/dl
Interpretation of the result
If the result was at Normal value, the interpretation will be :
This patient has Normal Level of uric acid.

If the result was Higher than normal value, the interpretation will be :
This patient has Hyperuricemia

If the result was Lower than normal value, the interpretation will be :
This patient has Hypouricemia
Clinical significance
Creatinine
Estimation
Creatine & Creatinine
Creatine :
Creatine is a nitrogenous organic acid, synthesized
primarily in the liver,
then transported to the muscle and converted to
creatine phosphate, where it serves as high-energy
source

Creatinine :
Creatinine is a chemical waste molecule that is
generated from muscle metabolism, and it is
produced from creatine.

Reference ranges :
Male : 0.9 – 1.3 mg/dl
Female : 0.6 – 1.1 mg/dl
Clinical correlation of Creatinine
Creatinine concentration Measurement :
Is used to determine the sufficiency of kidney function & severity of kidney damages.

Creatinine clearance test :

Is a test that compares the creatinine level in urine with its level in blood,
and it is used to assess the renal function.
