PcDNA

 Partial complementary single strand DNA or

 P-dominant CDNA or

 Plasmid control DNA vector or

 Protamine complementary DNA etc.

 PcDNA is DNA vector used to clone recombinant
DNA sequences for the expression of proteins of
interest in mammalian cells.
OR
 plasmid c-cytomegalovirus promoter DNA vector

that is able to express a protein of your interest in

mammalian cell culture.
 Plasmid vectors were used for construction of DNA
libraries.
 The colonies were sometimes pooled, amplitude, in

liquid culture, and stored at -80`c.

 Can also mainfained in immobilized form on

nitrocellulose membrane.
 Most common vectors include Psport 1,PCMU-script

and PcDNA.
 Various types of host-vector systems have been

designed to broaden the host range.

 Contain SV40 and polyoma origins a transcription
unit comprising the human CMV promoter.

 PcDNA vectors designed for over production of

recombinant proteins in mammalian cell lines.

 The human CMU promoter provides high-level

expression in a wide range of mammalian cells.
 PcDNA a vector containing 3.2kb fragment with
beta-galactosidase gene c-terminal peptide.
 To propayte and maintain the PcDNA vectors
resuspend the vectors in 20ml sterile water.
 Make up 1t to mg/ml stock solution.
 Store at -20`c
 Use the solution to transform a rec A, top10, top 10F
etc.
