INTRODUCTION:

 Types of culture media

1. Solid Media
2. Liquid Media
3. Semisolid Media
Solid Media

 Agar : Solidifying agent, Employed at 2%, No nutritive value, solidify at 42and

melt at 95 degree Celsius
1. SCDA
2. SCA
3. R2A
4. MA
5. CA
6. MSA
7. XLDA
 1.SCDA (Soya bean Casein Digest Agar):
 General pupose media.
 Wide range of Microorganism grow on this media.
 This media is reffered by pharmacopeia for FP analysis.
 EM of clean area and manufacturing facility.
 Used in lab related testing.
 SCA (Sabouraud Chloramphenicol Agar):
 Recommended for yeast and mold
 Low pH approximately 5.6 support fungi
 Chloramphenicol is broad spectrum antibiotic.
 Inhibit wide range of Gram negative as well as Gram positive bacteria.
 R2A Agar:
 Low nutrient containing media
 Stress or injured microorganism promote to grow.
 Used in water testing.
 It contain pyruvate, help to repair the microorganism.
 MA (MacConkeys Agar):
 It is selective medium for enteric microorganism.
 It contain ox bile salt , inhibit Gram positive bacteria.
 It also contain crystal violet stain
 Media used in FP analysis as well as in water analysis.
 CA (Cetrimide Agar):
 It is also selective media.
 Used to confirm the presence of Pseudomonas aerugiosa.
 It contain cetrimide which is quarternery ammonium salt.
 Inhibitory action on wide range.
 Used in FP and Water testing.
 MSA (Mannitol Salt Agar):
 Selective media for staphylococcus aureus.
 MSA containing 7.5% salt conc.
 Nacl acts is inhibitory to all other bacteria.
 Ferment mannitol to produce yellow colour.
 XLDA (Xylose Lysine Deoxycholate Agar):
 Selective for salmonella species.
 Mediun containing sodium thiosulphate and ferric ammonium citrate.
 Medium contain sulphur which utilise by only salmonella species.
 Sodium deoxycholate act as selective agent for salmonella and it is inhibitory
to Gram positive bacteria.
