Food Test 1 for non-reducing sugars
Procedure:
1. Put 2 ml of the sucrose in a test tube.
2. Add 2 drops of citric acid solution to the food sample.
3. Heat the mixture to boiling in a hot water bath.
4. Remove the solution as soon as it boils and let the solution cool.
5. Add 2 drops of sodium hydroxide solution to the above sample.
6. Add 2 drops of Benedict's solution to the food sample.
7. Heat the mixture in the water bath again and record your
observations.
Food Test 2 for Reducing Sugar
Procedure:
• Using a dropper, take a small quantity of Million's reagent.
• Add few drops of Million's reagent to the test tube containing egg
albumin.
• Wait for some time.
• Pink colour appears in the test tube, which indicates the presence of
protein.
Food Test 4: Fat - Sudan III stain –
cooking oil
Procedure:
• To a test tube, add equal parts of test liquid and water to fill about
half full.
• If testing more than one liquid, label each test tube with a marker.
• Add 3 drops of Sudan III stain to each test tube. Shake gently to mix.
• A red-stained oil layer will separate out and float on the water surface
if fat is present.
Paper Spot Test
Procedure:
• Take a peanut seed from the watch glass.
• Crush the peanut seed and rub it on a piece of white paper.
• Paper becomes translucent at the spot, which indicates the presence
of fat.
Fat test using ethanol: cooking oil
Process
• Add the food sample to 2 cm3 of ethanol, shake well.
• Allow to settle in a test tube rack for 2 minutes for food to dissolve in
ethanol.
• Empty any clear liquid into a test tube containing 2 cm3 of distilled
H2O.
• A MILKY-WHITE EMULSION is a positive result: lipid is present.
• If the mixture remains clear, there are no fats present in the sample
