THE STRUCTURE AND

FUNCTION OF THE LIGHT

MICROSCOPE.
OBJECTIVES.

 State the purpose ( role) of a light microscope.

 List at least 5 parts of the light microscope.
 State the functions of at least 5 parts of the light microscope.
 Label at least 5 parts on a diagram of the light microscope
 Manipulate correctly the light microscope
 Calculate magnification of objects viewed from a light
microscope.
Focus questions.
Give an estimate of the sizes of the sand
grains in millimeters?
How many sand grains are shown in the
picture, try to count?

What prevents you from knowing the size
or number of grains present?

Can you name an instrument that can be

used to see the sand grains more clearly?
Possible Answers.
Give an estimate of the sizes of the sand grains in
millimeters?
How many sand grains are shown in the picture, try to
count?
Answer: too many to count cannot see all of them.

What prevents you from knowing the size or number
of
grains present?
Answer: they are too small to see or count.
Can you name an instrument that can be used to see
the sand grains more clearly?
answer: magnifying lens.

DISCOVERING THE MICROSCOPE

 Objects that are tiny so that they cannot be

seen with the naked eye are viewed with an
instrument called a microscope.

 A microscope magnifies ( looks bigger) or

increase the size of tiny objects such as cells
so it can be seen by using lenses.
Look at the
diagram of
parts of the
microscope
and locate
and name the
parts of the
microscope

Diaphragm Controls light passing to slide
Stage and stage Holds the slide in place

Arm supports the tube
Coarse adjustments Moves stage up and down for coarse
focus
Fine focus Moves stage up and down a tiny amount
adjustments for fine focus.

PREPARING OF MATERIALS TO BE VIEWED
UNDER MICROSCOPE.

 Objects (cells) are placed on a slide and viewed under the

microscope.
 Microscope slides are rectangles of glass.
 A specimen is placed on the microscope slide and covered
by a much thinner piece of glass called a cover slip.
 The cover slip prevents the specimen ( cell) from drying out
and also protects the objective lens of the microscope.

microscope the object ( cell) have to be very
thin to allow light to pass through it.
 Dyes or stains are often added to make the
image easily seen.
 Example: cheek cells are often stained with
methylene blue. This makes the cells look
blue when viewed with microscope.
