● Principle: carbohydrates when treated with concentrated sulfuric acid undergo
dehydration to give furfural derivatives. ● Procedure: put 2ml of carbohydrate solution in clean and dry test tube. add 2 drops of ethanolic Alpha Naphthol (Molisch reagent) and mix, after mixing , amount of 2ml sulfuric acid is slowly added down the sides of the sloping test tube ,without mixing to from 2 layers. ● Inference ( results ) : A positive Molisch test indicates the appearance of a reddish – purple or purple ring at the junction of two fluids. ● Interpretation : Benedict’s test ● Principle: Benedict’s test is a quantitative test. The color formed depends upon the amount of reducing sugar present in the mixture. ● Procedure: put 5 ml of Benedict’s reagent and after that add 8 drops of carbohydrate solution . Leave to boil on the flame or in a boiling water bath for 2 min . Then let the solution cool down. ● Inference ( results ): A positive test with Benedict's reagent appears by changing its color changes from a clear blue to a brick-red precipitate. The color formed depends on the amount of reducing sugar in the mixture or if the test tube contains carbohydrates, this solution is orange- red, but if it does not contain any carbohydrates, the color of this solution does not change. ● Interpretation: Barfoed’s test
