CELL AND TISSUE

CULTURE
PREPARED BY:
CHRISTIAMAE BARRA, LPT/BIOLOGIST
RELIABILITY OF DATA
• Random error
• Mean
• Systematic error
Errors of Methods
Instrumental Factors
Error of Operation
ASSESSMENT OF ANALYSIS
• Precision-closeness of replicate measurements which can be associated with random
error of the method.
• Accuracy-closeness of the mean of replicate analyses to the true value of the sample.
• Sensitivity-ability to detect small amount of test substance.
• Specificity
• Quality Assurance-The attitude of staff is of vital importance and they must be
motivated to produce reliable data.
• Control Samples
CELL AND TISSUE CULTURE

• Is the general name for removal of cells, tissues, organs from an

animal or plant and their subsequent placement into artificial
environment conductive to growth
MICROBIAL CELL CULTURE
• is a method of multiplying microbial organisms by letting them
reproduce in predetermined culture media through the microbial
cell under controlled laboratory conditions
CULTURE AND MEDIA

• Media : Nutrients used to grow organisms outside of their

natural habitats
• Culture : The propagation of microorganisms using various
media
MAIN TYPES OF MEDIA (PHYSICAL STATE)

• Liquid
• Semisolid
• Solid
MAIN TYPES OF MEDIA (CHEMICAL
COMPOSITION)
• (I) natural or empirical cultural media
• (2) semi-synthetic media
• (3) synthetic or chemically defined culture media
• (4) living media
• (5) functional type general purpose
• (6) oxygen requirement
BASIC REQUIREMENTS OF CULTURE MEDIA

• Nutrients (Energy source, Carbon source and Nitrogen source)

• Mineral Salts (Sulphate, Phosphates, Chlorides, and carbonates of
K, Mg and Ca)
• pH (7.2-7.4)
• Water
• Growth factors
CULTURE MEDIA
• Food material on which microorganisms are grown in the laboratory
Chemolithotroph
Chemoorganotroph
Photolithotroph
Cell line
• Chemoautotroph would contain salts of Na+, K+, Ca+, Mg+, NH4or N03 -, CC
HP042, S042- and a simple carbon source such as glucose.
PLANT CELL AND TISSUE CULTURE
5 CLASSES BASED ON TYPE OF MATERIAL
USED
• Callus Culture
• Cell Culture
• Organ Culture
• Meristem Culture
• Protoplast Culture
ANIMAL CELL CULTURE

• Animal cells are derived from tissue explants or from disaggregated embryos.
• growth of tissues was possible in vitro in sterile medium containing salts,
carbohydrates, vitamins and amino acids, and serum supplements.
MAMMALIAN CELL CULTURE
• Directly from fresh tissue
• complex

Two types
• Primary Culture
• Cell lines
