Basic principles of

Genetic Engineering
BY: SHWETA SINGH CHAUHAN
ASSISTANT PROFESSOR
SRM UNIVERSITY
Introduction
Genetic Engineering primarily involves manipulation of Genetic material (D.N.A) to
achieve desired goal.
Other terms used in Genetic Engineering:
• Gene manipulation
• Recombinant DNA Technology
• Gene cloning
• Gene modification
• New Genetics
Principles of Genetic Engineering (R-DNA
Technology)
1. Generation of DNA fragments and selection of desired piece of DNA.
2. Insertion of selected DNA into cloning vector (e.g. plasmid) to create a recombinant
DNA or chimeric DNA.
3. Introduction of recombinant DNA into host cell (Bacteria).
4. Multiplication and selection of clones containing recombinant molecules.
5. Expression of the gene to produce the desired product.
Genetic Tools used for Recombinant DNA Technology

• Desired Gene
• Restriction Endonuclease enzyme (for cutting at particular site)
• Cloning vector
• Ligase enzyme (for Ligation of vector of DNA with desired piece of DNA)
• Host cell (Bacteria, Bacteriophage, Virus, Yeast)
• Recovery of Gene
