availability.

Biosafety considerations for research with
Lenti viral Vectors:
• Attractive features like convenience, high efficiency, Broad cell range,
stable integration.
• Such research also raises biosafety issues.
• NIH office of Biotechnology received frequent questions regarding
appropriate containment for lenti viral vectors.
• RAC (Recombinant DNA Advisory Committee) was asked to provide
additional guidance for institutional biosafety committees(IBSC) and
investigators.
• Risks of Lentivirus vectors:
• Potential for generation of replication competent lenti virus( RCL).
• Potential for Oncogenesis.
These risks can be mitigated by the nature of vector system and its
safety features.
• General containment considerations:
• Either BL-2 or advanced BL-2.
• Use of PPE to avoid mucosal exposure to the Vector.
• Animal studies:
• IBSC considers the biosafety issues associated with animal husbandry
and housing.
• Initial delivery of Vector need to be done in BSL-2 for animals. As to
minimize the risk of autoinoculation by the investigator.

• If there is no expectation on infection it is permissible to reduce the
containment from BSL-2N to BSL-1N, Within a few days specified by
the local IBC(1-7 days).

 G-II-B-3-a. Biological safety cabinets(class 1 and 2).
 G-II-B-III-a-(1). Procedures with a high potential for creating Aerosols.
 G-II-B-3-a-(2). Large volume of organisms containing recombinant
DNA molecules are used. May be centrifuged in the open laboratory if
sealed or centrifuge safety cups are used and if they are opened only
in Biological safety cabinet.

• Appendix G-II-B-4. Laboratory facilities (BL-2).
• G-II-B-4-a. The laboratory is designed so that it can be easily cleaned.
• G-II-B-4-b. Bench tops are imperious to water and resistant to acids,
alkali, organic solvents, moderate heat.
• G-II-B-4-c. Laboratory furniture should be sturdy and spaces between
benches, cabinets and equipments are accessible for cleaning.
• G-II-B-4-d. Each laboratory contains a sink for hand washing.
• G-II-B-4-e. If the laboratory has windows that open, they are fitted
with fly screens.
• G-II-B-4-f. An autoclave for decontaminating laboratory wastes is
available.
