TRANSMUCOSAL PERMEABILITY
TRANSMUCOSAL PERMEABILITY
• Transmucosal permeability refers to the transport of
material such as drug across the oral mucosa .
• Two routes are involved in drug permeation across epithelial
membranes
1. Paracellular route [aqueous pore pathway ]
2. Transcellular route [lipoidal pathway ]

1. Paracellular route
• Passage of molecule through intercellular space.
• Water soluble substances and ions probably move through
the intercellular space .
1. Transcellular route
• transport into and across cells .
• Substances with a high solubility in lipid are
expected to transverse the oral mucosa more
easily by moving along or across the lipid reach
plasma membrane of the epithelial cells.
FORMULATION CONSIDERATION OF BUCCAL DELIVERY SYSTEM
Basic components –
1. Drug
2. Bioadhesive polymers
3. Backing membranre
4. Plasticisers
5. Permeation enhancers
6. Preservatives

7. Drug – before the formulation it has to be decided which type of dosage


form required
• Rapid release
• Prolong release for local systemic effect
Characteristics-
1. Conventional single dose of drug should be small
2. 2-8 hour of drug half life should be selected
3. The drug undergoes passive absorption on oral administration
2. Bioadhesive polymers- bioadhesive play a major role in bdds
hence it should considered at initial covalent phase selection of
dosage form, characterization of polymer
Property-
• Should be inert and compatible with environment
• Should be non toxic and absorbable from the mucous layer
• Have high molecular weight and narrow distribution.
Ex.- agarose, gelatin,HPC HPMC.

3. Backing membrane- the outermost layer of patch which format


the formulation during wear period.
• The material used as backing membrane ex.carbopol,
magnesium stearate,cmc

4. Penetraion enhancer- effective in delivering high molecular


weight compound and exhibit low buccal absorption rate.
• Ex.-soudium lauryl sulphate polysorbate ,azone
5. plasticisers- these are used of achieving softness
and flexibility of thin flims of polymers . They also
act as a permeation enhancers
Ex.- glycerol,PEG200,PEG400
6.Preservatives- parabens , benzalkonium
chloride ,phenyl ethyl alcohol, EDTA are the
examole of some commonly used preservatives
