NINHYDRIN TEST
Test

for amino acids and proteins with a free NH2 group When an -NH2 group reacts with ninhydrin (0.1% triketohydrindene hydrate), a purple-blue complex is formed

OVER-ALL REACTION:

MECHANISM:

Amines

(including -amino acids) react with ninhydrin to give a colored product.


-amino acids typically give a blue-purple product (positive result). [Eq. 2] a secondary amine, gives a yellow-orange colored solution as a condensation product. [Eq. 3]

can be used qualitatively (e.g. for chromatographic visualization) or quantitatively (e.g. for peptide sequencing).
test is sensitive enough that ninhydrin can be used for the visualization of fingerprints.

PROCEDURE:
1 mL diluted plasma was put in a test tube and added with 0.5 mL of ninhydrin reagent. Solution was mixed, heated for 1 minute and allowed to cool. Colored solution formed was noted.
Ninhydrin test was repeated using 1 mL of 0.3% soution of glycine (positive control) instead of plasma. Color obtained was compared to the diluted plasma.
