Aim: To prepare a stained, temporary slide of an onion peel.
To observe it under microscope to record an observation.
Apparatus: A compound microscope, a sharp knife, a sharp blade, forcep, needle, glass slide, cover slip, petri dish,
dropper

• Material- an onion peel, magnifying glass, filter paper

 Procedure:

• Take an onion peel with tied thick fleshy leaf carefully.Use the forceps to gently
remove the transparent membrane (peel).

• Transfer it to distilled water and cut a smaller piece of a peel.Put a few drops of
methylene glue and add few drops of water to it.

• Use a brush to transfer the stained peel to petri containing water.After a minute
transfer the peel on the glass slide with the help of filter paper.

• Put a drop of water and cover the needle to remove the air bubble
Observation:

• A large membrane of cell are seen lying side by side.

• The cell have a distinct nucleus and vacuole in the cytoplasm.

• Shape of the cell- polygonal

• Cells are compactly arranged
