• Preparation of spawn for commercial


production of button mushroom :-
1. Spawn :-

• Spawn is a pure mycelial growth, grown on sterilized


cereal grains, straw or swit.
• Spawn is the seed of mushroom.
• The spawn (seed of mushroom) is a pure culture of the
mycelia grown on a special medium.
• The medium is prepared by the grains of wheat, rye,
sorghum or bajra along with some ingredients.

Master spawn :-

• The spawn which is prepared by inoculating sterilized


cereals grain or straw or sawdust pure culture is called as
master spawn.
• Master spawn is nothing but the mushroom fungus
grown on a grain based medium
• Pure culture raised either from tissue or spores is
inoculated in a suitable substratum,which provides food
to the mycelium

Commercial spawn :-

• Spawn prepared by inoculating sterilized cereal grains,


straw or sawdust with master spawn.
• Master spawn or master culture bottles are further used for
inoculation of large number of other grain bags / bottles
prepared by the same technique and resultant is the
commercial spawn

• Steps for Preparation of Spawn for


commercial button mushroom production:-
1. Select unbroken, not very old & clean sorghum or wheat grains.

2. Boil the grains in water. Take precaution that the grains don’t
split to give out starch & they don’t become too soft.

3. Remove excess water using aluminium mesh & spread the


grains over jute cloth.

4. Then add 2% CaCO3 & 2% gypsum OR 3-4% CaCO3 alone to


the grain on wet weight basis. Mix it well & fill it in polythene
bags of suitable size.

5. Then plug this bags tightly & sterilize it @20lbs/psi for 2


consequent days

6. Now, inoculate the sterilised bottles with pure culture and


inoculate at 24+ - 2°C for 2 weeks

7. In this time the mycelial growth covers the grains &


conspicuous whitish thread like growth is observed on the grains.

8. Commercial spawn can be prepared by inoculating 20


polypropylene bags from 1 bottle of master spawn

• The process of mixing spawn with compost is called


Spawning.

A. Tray system of cultivation:- The compost when


ready is taken in trays leaving about 5 cm from the top.

• Single layer spawning:-


The grain spawn is scattered
uniformly all over the compost surface in a tray, which is
then covered with a thin layer of compost 200g/m bed
spawn.

• Double layer spawning:-


The trays are half filled &
spawn is scattered. Then the tray is filled & completely
compost & spawn is applied as above & covered with a
thin layer of spawn.

• Through spawning/mixed spawning:-


The required
quantity of spawn is mixed with the compost & filled in
trays or polybags.

• Spot spawning:-
The compost is filled in trays &
holes of 2.5-5cm deep are made with fingers about 8-
12cm apart in rows. About 5gm spawn is introduced in
to cavities & covered with a thin layer of compost.
After spawning, cover the beds with sterilized
newspaper & sprinkle water over the papers to maintain
moisture level of compost
The temperature should be around 25±2°C with relative
humidity of 85-90%.

B) Bag system of cultivation:-

1. In this method polythene covers of size 35“x 24" with


150 gauge thickness are used.
2. The spawn is mixed with the compost @
500-750gm/100kg compost.
3. Then mixed compost is filled in polythene bags @ 15-
20kg/bag.
4. Finally, the beds are covered with newspaper.
