This study aimed at analyzing the effect of Pb exposure towards pupation

and imago behaviors of D. melanogaster
Methodology
The strain used in this research was D. melanogaster
wild-type.

There are four Pb levels given, which were 0, 1000, 1500,

and 2000 ppm.

The data collected were consists four behavioral assays such

us pupa position, pupa orientation, the success of imago
climbing, and the period needed by imago to climb

The data collection was done in generation 0 and generation

1. The data analysis used were percentage (pupation) and
one-way ANCOVA (climbing ability) followed by the LSD
Test
Pupation Assay

3 Place the newly Flies are allowed to calculate the number of distribution of pupae
hatched transplanted mate for 9 days in each zone, and the number of positions of
flies in the test bottle (until pupae appear) the pupa (upright, tilted, flat) in each zone

Climbing Assay

Ability : Number of flies that can

successfully climb at a height of
The imago is inserted into the test tube Tap the tube until 8cm in 10 seconds
Climbing ability : 10 tails all imago are at the Time : Average of the length of
Length of time climbing : 5 tails bottom of the tube time it takes a fly to climb to
reach a height of 8cm
Result dan Discussion
The data analysis percentage of pupation show that Tilted was the majority
position of pupa in each treatment either in generation 0 or generation 1. The
analysis results of pupa position showed that the lower the zone the higher the
number the pupa found.

Note :
1. A: control, B: 1000ppm, C: 1500ppm, D: 2000ppm
2. Upright position: pupa position with an inclination of 0o / 180o .
Position, Tilted: position of the pupa with a slope of 45 o / 225o ,
flat position: the position of the pupa with a slope of 90 o / 270o .
3. Zone 1 (very low), zone 2 (low), zone 3 (high), and zone 4 (very
high)
Furthermore, Pb treatment has significantly affect the success of climbing [F(3,19) =
34.132, p < 0.05] and climbing period [F(3,19) = 26.604, p < 0.05] of Drosophila
imago.

In conclusion, Pb affect the behavioral changes of D. melanogaster.

The possible factor is that the nerve condition which is contaminated by Pb will result
in disruption of the work of the brain of D. melanogaster and will affect the behavior,
one of which is climbing, because the brain will capture all stimuli to be understood
with the help of the activity of neural circuits, nerve cells and neurotransmitters.
Disrupted brain work will affect behavior, because brain activity raises a behavioral
response in an individual (Ramdya, Schneider, & Levine, 2017).
