 Cholesterol : An Overview

• Cholesterol is unsaturated steroid alcohol containing : four rings AND C-H chain side just like fatty
acids
• hydrophilic side : hydroxyl group in A ring
hydrophobic side : the rest of its structure
• it makes cholesterol can both interact with lipid and water
• cholesterol is mostly synthesized by animals
• unlike other lipids, cholesterol isn’t readily catabolized by cells. Otherwise can be converted into
primary bile acids eg colic acid and chenodeoxycholic acid.
• both colic acid and chenodeoxycolic acid help the absorption of fat in intestine by acting like
detergent.

• Cholesterol can be converted by adrenal gland, testis, ovary, steroid hormones.

• Can be converted into vitamin D by irradiation of sunlight

• The early lipid workup has begun with measurement of total serum chol
• past decade : using strong acids (H2SO4 and acetate acid) together with other chemicals (acetic
anhydride or FeCl3) to produce colored chol.
• present decade : enzymatic reagents are used.
• enzymes are selected because their specificity to the analytes and lead to accurate quantitation.
• all types of chemical lipids (HDL, LDL, chol, and lipoprotein) are measured from the chol content
