Extraction and Purification of Protease from

Culantro, “Awa-phadigom” in Manipur.

• Introduction

Culantro (Eryngium foetidum), local name Awa-phadigom, is a binneil medicinal leafy green
herb belongs to the family Apiaceae. It is commonly grown in moist and shaded areas.
It is found in West Indies and several countries in South America and Asia. In India, it is
found mostly in North-East state of Sakkim (Bhotay dhania), Assam (Man dhania), Tripura
(Bilati dhania), Nagaland (Burma dhania) (Tshering Tashi Lepcha et al., 2018).
Culantro content various nutreints, moisture (83.3%), crude protein (2.63%), sugar (8.26%),
ascorbic acid (32.3 mg/100g), fat (0.73%), ash (3.0 %) (Tshering Tashi Lepcha et al., 2018).
Protease can hydrolyze protein and its activity is already reported from cilantro (coriander).
Culantro is widely used in seasoning a large vareity of foods that includes curries, meats,
soups, etc.
Medicinal uses of culantro:
Treatment of infectious disease
Treatment of diabetes
Treats fever
Detoxification

Materials and Methods

Materials
Beakers, reagent bottles, conical flasks, plastic bottle , wash bottle, measuring
cylinder of various capacities, centrifuge and digital balance.
Extraction buffer( 0.125M tris HCl), casein, TCA, ammonium sulphate.
Collection of sample : The fresh sample along with roots collect from From
districts of Manipur.
Sample preparation: Leaves are removed, cut and washed.
