In 1987, the federal government approved the


establishment of the National institute for Pharmaceutical
Research and Development (NIPRID) as a parastatal under the
Federal Ministry of Science and Technology. This approval was
based on the recommendation of the pharmaceutical society of
Nigeria (PSN).

INTRODUCTION TO PACKED CELL VOLUME

The Packed Cell Volume (PCV) test, also known as


hematocrit or crit value, is a diagnostic test used to measure
the percentage of red blood cells (RBCs) in a given blood
samples. This test is useful in determining the concentration
of RBCs in the blood and can help diagnose anemia or
dehydration.
APPARATUS USED FOR PCV TEST
Hematocrit Reader: is used to measure the percentage of red blood cells
(RBCs) in a patient blood.

Micro Hematocrit Centrifuge: spins a small volume of blood in a capillary


tube to separate the cellular components from plasma, allowing for the
measurement of the hematocrit level.
APPARATUS USED FOR PCV TEST

Centrifuge Hematocrit tube: this is a small, specialized tube used to separate


blood components by centrifugation. The hematocrit tube is used specifically
to measure the percentage of red blood cells in the blood, which is known as
the hematocrit.
METHOD OF PCV TEST

 Collect the blood sample from EDTA bottle


 The heparinized capillary tube will be filled to 2/3 length from the blood sample, before it
will heated on flame using Bunsen burner.
 Then, the absorbent cotton wool is used to clean the tube before it is placed in the
centrifuge.
 Placement of the sealed tube into a micro-hematocrit centrifuge machine.
 The seal must touch the outward base of the spinner.

 Place the seal tube in a hematocrit centrifuge at 12,000/13,000rpm for 5mins.


 The tube is now placed on the micro hematocrit reader to read the result in percentage.
 Position the slot so that the base line intersect the base of red cells and also move the
tube holder so that the top line intersect meets the top of plasma.
 Adjust the knob so that it meet the middle line intersect of the red cell.
PERCENTAGE RANGE OF PCV TEST

The Percentage Range of PCV test are:


PERCENTAGE
RESULT ADULT: NORMAL RANGE FOR MALE 37-50%
NORMAL RANGE FOR FEMALE 35-45%

CHILDREN: 2-5 YEAR 34-46%


6-12 YEAR 35-45%
A decrease in PCV indicates ANAEMIA while an increase in PCV indicates
POLYCYTHAEMIA
CAUSES OF LOW AND HIGH PCV

Low PCV:

 Blood loss
 Nutritional deficiencies
 Chronic diseases

High PCV:

 Low oxygen levels


 Smoking
 Inherited disorders
 Kidney diseases
ADVANTAGES OF PCV

It is used in the diagnosis and treatment of


anaemia.
It helps in the diagnosis and treatment of
polycythaemia.
It helps in the determination of extent of
dehydration and recovery from dehydration after
treatment.
It is helps in the decision of blood transfusion.
FACTORS THAT AFFECT PCV TEST

 Interruption in power supply.


 Wrong or poor blood sample collection.
 Parallax error while reading the result on the hematocrit reader
 Incorrect blood to anticoagulant ratio
 Over spinning of the blood in the centrifuge, lysing of the blood by flame or delay in
spinning.
CONCLUSION

The PCV test is an essential diagnostic tool


used in clinical practice to aid in the
management of patients with blood disorders.
