Quarter 1 Module 6 to 7
 Learning Competency
 : Describe the biological
techniques, procedures and
methods
Lesson Objectives
 1. Describe some of the aseptic
techniques used in the laboratory
 2. Identify the applications of
these techniques in one’s own
home 3. Give the importance of
practicing these techniques in the
laboratory and at home

Let’s Review
 What are some of the laboratory
equipments used in
Biotechnology? Give one and
state its function.
 Why is it essential to understand
the uses and application of these
equipments?
Engagement

 Interpret

If life may seem to trick

you and give you the boot,
there is only one that can lick
you and that is the guy inside
your suit.
Engagement
 In this time of pandemic, what are the
routines you do at home to get rid of being
infected by the Covid19?

 In preparing foods, how will you make sure

that your food will not be easily spoiled?
Let us learn together.
 Aseptic technique is a set of routine
procedures to ensure that culture media,
solutions and other equipment used in the
laboratory remain sterile and not
contaminated with other microorganisms.
Same is done in the field of medicine and
surgery to prevent infection of wounds.
 Ifthe biologist wants to grow a bacteria
in a culture medium it would be a
mistake if other microorganisms were to
grow on the culture medium. Aseptic
techniques are employed to prevent
such contaminations. These techniques
minimize the chances of other
microorganisms from the environment to
grow on the culture, as well as the
chances of the specimen contaminating
the environment.
 The study of microorganisms is one of the
exciting areas in biology. Because of this,
methods of cultivating and growing
microorganisms have been a mainstay in many
studies. These include growing them, identifying
them as well as which antibiotic or chemical will
be able to prevent their growth. Since the
microorganisms are living things, they need one
thing to survive and that is the source of energy.
Humans and animals eat food to get our
energy.
 Plantsuse photosynthesis to make
their own food. Microorganisms such
as bacteria are not different. They
also need a source of energy which
we need to provide if we are to grow
them in a culture. In the laboratory,
we use culture media as a tool to
grow or culture microorganisms.
 Culture media are different materials used
to grow or culture microorganisms. They can
be liquid or gel in consistency. The most
culture media are called nutrient broth, and
agar plate. Depending on the organisms you
wish to grow, your choice of medium maybe a
nutrient broth composed of a peptone with beef
extract. These provide the microorganisms
with amino acids, fatty acids, carbohydrates,
salts and other compounds for their growth.
Aseptic Techniques
Cleaning and disinfecting lab
surfaces prior to use

Limiting the duration that

cultures or media are uncapped
and exposed to the air
Keepingpetri dishes closed
whenever possible

Effectively
sterilizing inoculating
loops and other equipment that
comes into contact with cultures
or media

Avoiding breathing on cultures

or sterile instruments.
 Q1. What are common cleaning and disinfecting
agents are used in your home? Do you know how
they work?

 Q2. Why do we need to cover our food at home?

What will happen if we let it uncovered?

 Q3. Why should you avoid breathing on cultures or

sterile instruments?

 Q4. What are the reasons why we cook food?
