Chromatography

Introduction

 Chromatography is a separation technique which

is used for separation of mixture into its
components.
 The discovery of chromatography is generally
attributed to the Russian botanist Mikhail S. Tsvet
(also spelled Tswett).
 Chromatography may be preparative or analytical.
Uses
 Pharmaceutical Industry
 Diagnostic labs
 Forensic labs
 Food Industry
Pharmaceutical
Industry
Forensic Lab

To identify samples of drugs,

explosives, inks and
biological samples like saliva,
urine and blood etc
Food Industry

• Identification of
components
• Analyzing the products
• Testing the products on
the basis of nutritional
quality.
Why we use in cell biology?
Components of chromatography

 Mobile phase
 Stationery phase
 Sample mixture to be analyzed
 Principle of chromatography
Types

 Paper chromatography
 HPLC chromatography
 GC chromatography
 GLC Chromatography
 TLC chromatography
Paper
Chromatography
The principle of paper
chromatography is based
on the varying degree of
interaction between the
constituents and the
stationary phase.
What is
RF Value?

The Rf (retardation
factor) value is the ratio
of the solute's distance
travelled to the solvent's
distance travelled.
Applications

 Used for separation of mixture having polar and non

polar compounds.
 For separation of Amino acids.
 Used to determine organic biochemical in urine etc.
 Forthe evolution of inorganic compounds like salt and
complex
 Used in pharmaceuticals and food industry.
