Colony Forming Unit

CFU
Microorganism and Us
• Microorganism play an important role in our lives. Some of them are
beneficial in many ways whereas some others are harmful and cause
disease.
• Microorganism may be single celled like bacteria, some algae and
protozoa, or multicellular like many algae and fungi.
• Microorganism may exist in alone or colonies.
Microbial contamination of DHA oil
• Microorganisms are ubiquitous and diverse and thus can be found in a diversity of
environments thriving under varying conditions. Algae oil is a rich source of carbon, it
provides a suitable medium for microorganisms to exist and thrive under the right
conditions of pH, temperature, aeration, and pressure.
• The findings of this study indicate that there was seemingly high Coliform
contamination in oil samples.
• Coliforms, broadly classified into lactose fermenters and non-lactose fermenters, are a
diverse group of microorganisms with some associated degree of pathogenicity and
toxicity, particularly with some species such as Salmonella, E.coli, Pseudomonas,
Klebsiella, etc.,
Colony forming unit (CFU)
• As part of daily routine, in industrial Laboratory has to determine the number of
bacteria in a given sample as well as having to compare the amount of bacterial
growth under various condition.
• Enumeration of microorganism is very important in dairy microbiology, food
microbiology and water microbiology.
• Microorganism are so small and numerous, counting them directly can be very
difficult. Some of the methods used involve diluting the sample at which the
number of microorganism has to reduced, this enable an estimate to be
established for quantifying the microbes.

Five gram of oil sample was weighed and One gram of oil sample was weighted
45ml of peptone water measured and added and 100ml Tween 80 was measured and
to the weighed sample and mixed. added to the weighed sample and mixed.

To help to reduce the dense culture of cells concentration, it was serially diluted to 10-¹, 10-², 10-³ and 10-

Tryptic Soya Agar
Non-selective media providing enough nutrients to allow for a wide variety
of microorganisms to grow.
• Colony characteristic: *yellow colonies surrounded by yellow zone.
*Pink colonies and red colonies
*Large white colonies
• Form: * Circular * Irregular

Eosin-methylene blue Agar
EMB is a selective, differential agar medium used for isolation of gram
negative rods in a variety of specimen types.
Colony characteristic > Dark purple

Mannitol Salt Agar
• Mannitol Salt Agar (MSA) is used as a selective and differential medium
for the isolation and identification of Staphylococcus aureus.
• Colony characteristic > Pink colonies
• Form > Circular
• Elevation > Convex
