Total white blood cells count

Total White Blood Cells Count:
 WBCs count and Differential count are one of
the most common and routine laboratory tests,
and it is one of the first steps in diagnosing
prevalent an illness in the Sudan .
 Calculations of A WBC is performed with
hemacytometer.
 Used a glacial acetic acid to count WBCs.
Principle:

Whole blood is diluted by mixing


with a fluid that lyses the red
cells, dilute the white cells and
stains its nuclei deep violet-
black.
 Reagents and
Equipment
 glacial acetic acid
 EDTA tube
 1ml pipette ,0.02ml
pipette and Pastter pipette.
 Hemacytometer.
 Test tubes.
 cover glass.
 Petri dish.
 Microscope.
 Cotton and Gauze.
glacial acetic acid useful for :
 dilution of blood.
 lyses the red cells.
 white cells remain intact and their nuclei
staining deep violet.
Method:

 Add 0.02ml of blood to 0.38ml of diluting fluid in a


glass tube (dilution 1 in 20).
 Mix well for at least 1 min.
 Prepare the counting chamber (Clean, dry and fix the
cover slip).
 Smoothly fill the chamber.
 Leave the chamber undisturbed on a bench for at
least 2 min for the cell to settle.
 Using X 10 objective, count the white cells in the 4
corner of the chamber.
 Calculate the WBC cells/mm3 =

Example :
 Sq1 (20) cells, sq2 (30)cells, sq3 (28)cells,
sq4(25)cells.
 WBCs count =

 5,150 cells/mm3
Normal Ranges:
 Adult: 4,800 to 10,800 white blood
cells/mm3
Newborn: 10,000 to 30,000 white blood
cells/mm3
