Transdermal Drug Delivery System

 Transdermal drug delivery system

 Drug absorption via the skin.

 Improve the therapeutic efficacy and safety of the drugs
A new form of delivery system called the microneedles
 To enhance the delivery of the drug through this route and
overcoming the various problems associated with the
conventional formulations.
MICRONEEDLES
A device consists of polymeric needles of micron size,
which are arranged on a small patch to deliver sufficient
amount of drug to give a required therapeutic response.
Comparison of topical cream, hypodermic needle, microneedle patch
and transdermal patch.
AIM OF Micro needle DDS

Onset of action
Faster

Pain Painless

Bioavailability Sufficient

Patient compliance Better

Self-administration Possible

Mechanism of drug delivery(Diffusion Bypass stratum corneum and drug placed directly into
Mechanism) epidermis or dermis hence enhanced permeability
Mechanism of drug delivery

(1) Microneedle device with drug solution

(2) Device inserted into the skin
(3) Temporary mechanical disruption of the
skin
(4) Releasing the drug in the epidermis
(5) Transport of drug to the site of action.

DIMENSIONS OF MICRONEEDLES

❖ Needle length of up to 1500 μ is sufficient

to release the drug into the epidermis.
❖ Mostly they are 150–1500 μ long
❖ 50–250 μ wide
❖ 1–25 μ tip thickness.
❖ SHAPES: cylindrical, triangular,
pointed, pentagonal, octagonal
TYPES OF MICRONEEDLES

Fabrication Material:

1.SILICON
2.METAL: stainless steel, titanium,
nickel, iron
3.SILICA GLASS: Borosilicate glass,
boron trioxide
4.POLYMERS : poly lactic acid(PLA),
poly glycolic acid(PGA)
5.Ceramic : Alumina (Al2O3), Ormocer
Microneedles: A smart approach and increasing potential for
transdermal drug delivery system

Fabrication
 Microneedles can be fabricated employing micro electro mechanical system(MEMS)
 Basic process is divided into 3 parts

1. Deposition
2. Patterning
3. Etching
Methodology for drug delivery
 poke with patch approach
 Coat and poke approach
 Biodegradable micro needles
 Hollow micro needles
 Dip and scrape
Evaluation of microneedles

1. Characterization methods
characterizations including
particle size
polydispersity index
 viscosity
zeta potential
The size, internal structure, and crystallinity of the liposomes or nanocarriers can
be performed using a dynamic light scattering, X-ray scattering, and transmission
electron microscopy technique.
2. Dimensional evaluation:
(SEM)
Confocal laser microscope
