What is shoot tip culture
• Shoot tip culture may be described as the culture of terminal (0.1-1.0 mm) portion of a
shoot comprising the meristem (0.05-0.1 mm) together with primordial and developing
leaves and adjacent stem tissue.
Why shoot tip Culture

➤No cell differentiation
➤ virus cant move from cell to cell
➤Intense metabolic activity
➤No vascular system
PROTOCOL

Sterilization: Clean and sterilize shoot tips.


Prepare Medium: Create a sterile nutrient medium.
Initiate Culture: Transfer sterilized shoot tips onto the medium.
Incubate: Grow cultures in controlled conditions.
Subculture: Transfer growing shoots to fresh medium periodically.
Plantlet Development: Transfer developed shoots to soil.
1. Hardening Off: Gradually expose plantlets to outdoor conditions.
STAGES OF SHOOT TIP CULTURE
Stage 1 is the culture establishment stage
• when explant may develop into single shoot or multiple shoots.
• At this stage explant are supplements with cytokinin like BA, kinetin and
2iP.
In stage 2
• the objective is to multiply the propagule and for this axillary shoot
proliferation is followed as it maintains higher genetic stability.
The stage 3
• purpose is regeneration of adventitious roots from the shoots obtain in
stage 2
• Numerous studies have indicated that NAA is followed by IBA, IAA,
2,4-D and other auxins are used for induction of root generation
Factors affecting Shoot tip culture

Genotype: Different plant species and varieties respond differently to shoot tip culture techniques.
Nutrient Medium: The composition of the growth medium, including types and concentrations of nutrients,
hormones, and supplements, affects growth and development.
Hormones: Growth regulators such as auxins and cytokinins are crucial for initiating and promoting shoot
growth in culture.
Temperature: Optimal temperature ranges promote tissue growth and development while extreme
temperatures can inhibit growth or cause tissue damage.
Light: Proper light intensity and quality are essential for photosynthesis and regulating plant growth and
development in culture.
pH: The pH level of the growth medium affects nutrient availability and metabolic processes in the cultured
tissues.
APPLICATION

Propagation: Rapid multiplication of disease-free plants from a small


amount of starting material.
Germplasm Conservation: Preservation of elite plant varieties and
endangered species.
Genetic Transformation: Introduction of desired traits or genes into plants
for crop improvement.
• Disease Elimination: Production of pathogen-free plants through shoot tip
culture techniques.
